The Glenrothes distillery is located south of the town of Rothes. The Burn of Rothes, which originates in the Mannoch Hills, flows right through the grounds of the distillery. "Rothes" loosely translates as "meeting of the waters". The Burn of Rothes "meets" the "waters" of the River Spey northeast of the town of Rothes. Glenrothes indicates the place where the distillery was built, namely in the valley of the Burn of Rothes. The malt of Glenrothes is very popular with connoisseurs.

The distillery was built in 1878. On 15 May 1922, 200,000 gallons of whisky were lost in a fire in warehouse No. 1. In 1989, the stillhouse was completely renovated and the number of stills was expanded, after which two more stills were added in 1989. There are now five wash stills and five spirit stills in the Glenrothes stillhouse, the twenty washbacks are made of Oregon pine. For maturation, both refill casks and ex bourbon and sherry casks are used. For restoring the casks, the distillery has its own cooperage. Around the distillery are old lands of the Brauchhill farm. On these lands are four springs, which are owned by the distillery. The brewing water is drawn from the spring called The Lady's Well via a pipeline. To cool the condensers, the water is pumped from the small river Dounie Burn. Glenrothes' much loved malt comes as a single malt in a transparent belly bottle with a handwritten narrow label with the year.
